Protein-mediated uptake of vitamin B12 by isolated mitochondria.

Protein-mediated B12 uptake by isolated rat liver mitochondria has been shown to be enhanced by plasma transcobalamin (TC-II) but not by salivary R binder in vitro. The process is enhanced by calcium and depends on active mitochondrial respiration. Following uptake, cyanocobalamin is converted to adenosyl and methylcobalamins and released from the mitochondria. TC-II appears to be required for ...

Mechanism of intrinsic factor action in everted sacs of rat small intestine.

Three decades have passed since Castle (1) and his associates demonstrated the existence of intrinsic factor, yet knowledge of its mechanism of action is still so limited that in 1958 two workers felt justified in concluding there was no such thing as intrinsic factor (2). In prior studies (3, 4), a mechanism involved in the enhanced uptake of vitamin B12 by rat liver slices in the presence of ...

The role of methionine and vitamin B12 in folate incorporation by rat liver.

The decreased folate uptake induced by exposure of rats to nitrous oxide can be partly reversed by methionine supplementation. This is consistent with the theory that N2O inhibits methylcobalamin-dependent methionine synthetase (EC 2.1.1.13) in rat liver and previous observations showing decreased folate incorporation in rats made nutritionally vitamin B12 deficient.

Mechanism of vitamin B12 uptake by erythocytes.

Pitney, Beard, and Van Loon (2) and Ostrowski, Skarzynski, and Zak (3) reported in 1954 that vitamin B12 is bound primarily to a-globulin in normal serum. Pitney and his colleagues (2) also noted that p-globulin can bind B12 added in vitro, but they considered this as "free B12" because, unlike B12 bound to a-globulin, Euglena gracilis is able to utilize it without prior heat treatment. Miller ...

Gluconeogenesis from propionate in kidney and liver of the vitamin B12-deficient rat.

1. Kidney-cortex slices and the perfused livers of vitamin B(12)-deficient rats removed propionate from the incubation and perfusion media at 33 and 17% respectively of the rates found with tissues from rats receiving either a normal or a vitamin B(12)-supplemented diet. There was a corresponding fall in the rates of glucose synthesis from propionate in both tissues. 2. The addition of hydroxoc...
